Fluorescein diacetate (FDA) is a cell-permeable fluorogenic dye that is commonly used in various biological and biomedical applications. It is a non-fluorescent compound that can be converted to a fluorescent form inside living cells by cellular esterases. It is modified by acetyl groups, which enhance its membrane permeability and allow it to pass through cell membranes

Applications:
Viability Assays: FDA is commonly used to assess cell viability and determine the metabolic activity of living cells. It measures the presence of esterase activity, which is an indicator of cellular viability. The non-fluorescent FDA is added to the cells, and viable cells with active esterases convert it into the fluorescent form. The fluorescence intensity can be quantified using fluorescence microscopy or flow cytometry.Assessment of Cell Membrane Integrity: FDA is also used to evaluate cell membrane integrity. Intact cell membranes prevent the entry of FDA, and thus, only viable cells with intact membranes can convert FDA into its fluorescent form. Loss of membrane integrity is indicated by reduced fluorescence intensity.Microbial Staining: FDA can be used to stain bacteria, fungi, and other microorganisms. It is particularly useful for assessing microbial viability and differentiating between live and dead cells in microbiology and environmental monitoring.Assessment of Drug Effects: FDA is used in pharmacological and toxicological studies to evaluate the effects of drugs or compounds on cell viability and metabolism. It allows researchers to assess the impact of treatments on cellular health and determine the cytotoxic or cytostatic effects of various substances.Plant Research: FDA is used in plant biology research to study cellular viability and assess the health of plant tissues. It can be employed to evaluate the effects of environmental factors, such as stress, toxins, or treatments, on plant cells and tissues.
